PROBLEM TO BE SOLVED: To provide an operation method of a fluid handling device capable of simply and still more efficiently refining, recovering and inspecting a specific substance becoming an evaluation target. SOLUTION: The mixed solution 33 containing the specific substance in a second housing part 22 is only repeatedly sucked and discharged by the pipette 32 inserted in the housing part 22 to allow the mixed solution 33 in a first housing part 21 to repeatedly forcibly flow in both forward and opposite directions. By this method, the mixed solution 33 can be uniformly brought into contact with the surfaces of beads 30 being particles and the specific substance can be simply and efficiently captured by the beads 30.
