PROBLEM TO BE SOLVED: To provide a method for examining the presence and quantity of nucleic acid fragments having a specific base sequence such as difference in the poly A length and the repeat number of a repetitive sequence of e.g. microsatellite, single nucleotide polymorphism and insertion or deletion of base sequences, and to provide a genetic diagnosis to use the method. SOLUTION: The nucleic acid analyzing method includes the hybridization of at least two probes to a nucleic acid fragment, the binding of these two or more probes with a ligase, the conversion of pyrophosphoric acid formed by the binding reaction to ATP and the detection of chemiluminescence reaction depending on the ATP.
