1312631 Unsaturated 22-guanidino-steroids SCHERING AG 30 April 1970 [30 April 1969] 20797/70 Heading C2U Novel steroids of the formula and their acid-addition salts, which must contain at least one double bond selected from #<SP>1</SP>, #<SP>2</SP>, #<SP>3</SP>, #<SP>4</SP>, #<SP>5</SP>, #<SP>6</SP>, #<SP>7</SP> and #<SP>17(20)</SP> double bonds and which can contain further double bonds in the C and D rings (including #<SP>8</SP>), and in which X is CH 2 , CO, CH(α-OR) or CH(#-OR) wherein R is H or acyl, a 5-hydrogen atom when present has α- or #-configuration, the side chain at C-20 has R- or S- configuration and when a #<SP>17(20)</SP> double bond is absent contain H atoms on the 17- and 20-carbon atoms, are prepared from the corresponding 22-amino steroids or salts thereof and an S-(alkyl or aralkyl)-isothiourea or a salt thereof or cyanamide, or from the corresponding 22-alkyl- or aryl-sulphonyloxy steroids and a guanidinium salt, with, if desired, subsequent conversion of a resulting salt to the free base or conversion of a resulting free base to a salt. 22 - Amino - 23,24 - dinor - 5α - chola - 2,17(20)- diene is prepared from 3#-hydroxy-5α-pregn-17- (20)-ene-20-carbonitrile (prepared by hydrolysis of the 3-acetate) by conversion to the 3-tosylate, elimination of p-toluenesulphonic acid to give 5α - pregna - 2,17(20) - diene - 20 - carbonitrile and reduction of this. The novel steroids, which are stated to be active against bacteria, fungi and trichomonades, may be made up into pharmaceutical compositions with suitable carriers. |
