Provided are new 2,4-diamino-pyrimidine derivatives useful as aurora Ser/Thr kinase inhibitors. Further provided are methods of their use in the manufacture of medicaments to treatment and/or prevention of cancer, infections, inflammatory and autoimmune diseases. Also provided are combinations of the compounds with various cytostatic or cytotoxic cancer drugs.
