LEUCONOSTOC LACKING DEXTRANSUCRASE AND VECTOR FOR MAKING AND USING THE SAME

PURPOSE: A method for preparing mutant strain in which dextransucrase lacks in Leuconostoc is provided to stably express leuconostoc and improve fermentation food as a starter strain. CONSTITUTION: A method for preparing a vector for lacking a gene encoding dextransucrase of Leuconostoc comprises: a step of extracting chromosomal DNA from the Leuconostoc; a step of performing PCR using a primer; a step of obtaining fragment 991bp(sequence number 4) containing ORF and 777bp(sequence number 3) of dextansucrase gene ORF(open reading frame); a step of treating the fragment of sequence numbers 3 and 4 with a restriction enzyme; a step of treating pLC4255 vector with the restriction enzyme; and a step of performing binding reacting the treated fragment and pLC4255 vector. A method for preparing mutant strain in which dextransucrase gene lacks; a step of transforming the Leuconostoc with a vector for lacking dextransucrase gene; and a step of sub-culturing the transformed strain in a medium having antibiotics.
