PURPOSE: A method for demethylating target chromatin loci using histone demethylation is provided to enhance demethylation function and specifically regulate gene expression. CONSTITUTION: A method for demethylation of a target chromatin loci comprises: a step of expressing fusion protein of histone demethylase by conjugating delivery protein to histone demethylase gene; and a step of introducing the prepared vector to cells. The histone demethylation enzyme is JHDM3A. The fusion protein comprises an amino acid sequence of sequence number 1 or 2. The histone demethylase is a polypeptide having amino acid sequence of sequence number 3 or 4 or fragment of the polypeptide. The delivery protein is HP1α, Oct4, HP1B, HP1γ, Sox2, Nanog, Lin28, RASSF1A, RB or p53.
