<p><P>PROBLEM TO BE SOLVED: To provide a systematic algorithm maintaining the high specificity to a mutated site, and simultaneously selecting base sequences of many kinds of oligonucleic acid probes so that the melting temperature of a formed complex may be a previously set temperature. <P>SOLUTION: A partial base sequence of a corresponding site at the probe side is optimized based on the mutated site of a detection target nucleic acid molecule and partial base sequences of the neighboring bases, and the combination of base numbers in the complementary regions formed at the upstream side and the downstream side of the corresponding site is regulated. The selectivity to the detection target nucleic acid molecule and the melting temperature of the formed complex are each properly controlled by applying the two-stage base sequence-designing method of the whole probe to enable the design of the probe set optimum to the target detection condition. <P>COPYRIGHT: (C)2010,JPO&INPIT</p> |
