Isolated polynucleotides derived from Pseudomonas putida encoding polypeptides having styrene monooxygenase activity. The polynucleotides are derived from the StyA and StyB (StyAB) genes from Pseudomonas putida CA-3, using error prone PCR-based in vitro evolution and comprise at least one nucleic acid substitution at position 262, 449 or 1752. The polypeptides encoded by these polynucleotides are useful as biocatalysts for the synthesis of oxygenated compounds, as they exhibit improved properties, such as enzymatic activity and enantiospecificity. Also claimed are polypeptides having styrene monooxygenase activity. Methods for obtaining oxygenated compounds are also provided, as are organisms that express the polypeptides. |
