PLASTIDIAL THIOREDOXINS: OVEREXPRESSION AND BIOTECHNOLOGICAL APPLICATIONS

Plastidial thioredoxins: overexpression and biotechnological applications. There are described genetic sequences of m and f chloroplast thioredoxins (Trx) of the species N. tabacum, and the method of cloning, expression in plastids and applications thereof. There are also presented plastid transformation vectors containing DNA molecules which encode Trx, hosts incorporating the same, especially E. coli, and, in particular, transgenic plants obtained with such vectors, together with the method of obtainment and application thereof in the overexpression of Trx in active soluble form in said plants and in increased production of starch. Said plastid vectors are additionally applied to increased plastid production of recombinant heterologous proteins, fused to Trx sequences, specifically human serum albumin (HSA) and cardiotrophin-1 (hCT-1). There are also described methods of obtainment of heterologous proteins coexpressed with Trx, biologically active and in native conformation; together with the obtainment of recombinant hCT1 in soluble form having increased bioactivity.
