Disclosed is a method of synthesizing spongosine, which comprises reacting 1-O-acetyl-2,3.5-tri-O-benzoyl-beta-D-ribofuranose with 2-methoxyadenine to form 9-(2',3',5'-tri-O-benzoyl-beta-D-ribofuranosyl)-2-methoxyadenine, then deprotecting the 9-(2',3',5'-tri-O-benzoyl-beta-D-ribofuranosyl)-2-methoxyadenine to form spongosine. Also disclosed is the use of 2,6-dichloropurine in the synthesis of spongosine.
