<P>PROBLEM TO BE SOLVED: To isolate a new insertion sequence that can be mutated or transduced on a chromosome through a control mechanism different from the conventional insertion sequence, to prepare a plasmid into which the new insertion sequence is integrated or to prepare a transformant of coryneform bacterium. <P>SOLUTION: A plasmid that bears sac B gene originating from Bacillus subtilis and is replicable in the coryneform bacteria, for example, plasmid pMVD is used, transformed and smeared on a plate including sucrose.
