摘要 <P>PROBLEM TO BE SOLVED: To provide a method for modifying a nucleotide chain by which not only 3' end of single-stranded nucleotide chain but also 3' end of either one nucleotide chain of double-stranded nucleotide can selectively simply be modified. <P>SOLUTION: The method for modifying the nucleotide chain comprises adding a nucleotide sequence containing a specific base to 3' end side of the nucleotide chain to be modified, treating the nucleotide containing the specific base with a splitting enzyme specific to the nucleotide containing the specific base, imparting a functional group having binding ability to a desired modifying substance to the 3'-end of the nucleotide chain to be modified and then binding the modifying substance to the 3'-end of the nucleotide chain having functional group. In this method, the desired nucleotide chain can simply be modified without accompanying degradation of nucleotide chain itself and without taking unnecessary base sequence therein by adequately selecting the nucleotide sequence added. Owing to this process, the desired nucleotide chain can be labeled or tagged with the modifying substance and the modifying substance can stably and rigidly be immobilized on a substrate, etc., as a linker. <P>COPYRIGHT: (C)2006,JPO&NCIPI
