METHOD FOR TRANSFORMATION PROTOCOL OF BASIDIOMYCETE FUNGUS, POLYPORUS BRUMALIS USING PROTOPLAST ISOLATION

PURPOSE: A transformation of P. brumalis through protoplast isolation is provided to improve REMI(restriction enzyme mediated integration) and produce transformant of P. brumalis having enhanced lignin lysis activity. CONSTITUTION: A method for transforming P. brumalis comprises: a step of performing protoplast isolation from P. brumalis mycelium; and a step of adding gene for transforming to the protoplast; and a step of adding cell lysis enzyme liquid to the mycelium of P. brumalis and shaking. The cell wall lysis enzyme liquid contains 0.1-1.0% of USUKizyme based on 0.6M sucrose.
