<P>PROBLEM TO BE SOLVED: To provide a method for preparing a nucleic acid library comprising a plurality of elements. <P>SOLUTION: Each element comprises a first and a third stretches of nucleotides having the same sequence, and comprises the sequence of a second stretch of nucleotides having different sequences among the elements. A first ligation product is formed from the first oligonucleotide and a first oligonucleotide library, and cut with a type IIS restriction enzyme. The shortened oligonucleotide after the cutting is removed, and a second ligation product is formed from the first ligation product after the cutting and a second oligonucleotide library, and cut with the type IIS restriction enzyme. The shortened oligonucleotide after the cutting is removed, and a third ligation product is formed from the second ligation product after cutting and a fourth oligonucleotide, and cut with the type IIS restriction enzyme.
