PROBLEM TO BE SOLVED: To provide a method of preparing a test sample containing biomolecules of sufficient amount for measurement from a biological tissue. SOLUTION: The preparation method for the test sample containing the biomolecules comprises: a first preparing step which comprises adding a first buffer solution to the tissue picked from a living body, pulverizing the tissue in the first buffer solution, separating the obtained first tissue pulverized liquid into a first soluble fraction and a first insoluble fraction, and recovering the first soluble fraction to obtain a first test sample; a step for freeze-pulverizing the first insoluble fraction; and a second preparing step which comprises adding a second buffer solution to the freeze-pulverized products obtained by freeze-pulverization and mixing the resultant, separating the obtained mixture into a second soluble fraction and a second insoluble fraction, and recovering the second soluble fraction to obtain a second test sample.
