Provided are reagents, chaotropic agents, kits and methods for isolating nucleic acids based on magnetic cellulose materials, in which the chaotropic agents comprise an alcohol and a matrix solution, the matrix solution further comprises lithium salt, sodium salt or potassium salt. The alcohol concentration is adjusted by the matrix solution to appropriate value so that the nucleic acids within the sample bind to the magnetic cellulose materials, thus the non-nucleic acid materials will be separated from the nucleic acids.
