<P>PROBLEM TO BE SOLVED: To provide a DNA sequence encoding proteins useful in cell synthesis of PHA (polyhydroxyalkanoic acids), and to provide a method for producing PHAs (polyhydroxyalkanoates). <P>SOLUTION: A genome fragment harboring the gene phaG was cloned by utilizing phenotypic complementation of Pseudomonas putida KT2440 mutants, defective in PHA synthesis via de novo fatty acid biosynthesis but not affected in PHA biosynthesis via fatty acid β-oxidation. The 885-bp phaG gene encodes a protein of 295 amino acids with a molecular weight of 33,876 Da. The transcriptional induction of phaG has been observed under conditions of the PHA synthesis via the de novo fatty acid biosynthesis whereas no induction has been detected under conditions which favor the PHA synthesis via fatty acid degradation by the β-oxidation. The phaG gene is useful for the production of the PHAs in bacteria and plants. <P>COPYRIGHT: (C)2010,JPO&INPIT |
