PROBLEM TO BE SOLVED: To provide a method for simply determining or detecting DNA. SOLUTION: The method for determining the content of methylated DNA in an objective DNA domain of a genome DNA contained in a specimen originated from an organism has: (1) the first process for digesting the DNA sample originated from the genome DNA contained in the specimen originated from the organism with a methylating sensitive restriction enzyme; (2) the second process for obtaining a methylated single strand DNA from the digested DNA sample obtained in the first process and then binding the single strand DNA to a fixed methylated DNA antibody to select the single strand DNA; and a process (the first preprocess) as the preprocess of main processes, for separating the single strand DNA selected from the second process, from the fixed methylated DNA antibody to obtain the single strand state DNA (normal strand), and the like.
