Defective influenza virus particles incapable of producing functional polymerase

The disclosure generally relates to the field of influenza virus and vaccination against flu. The disclosure provides a method for obtaining a conditionally defective influenza virus particle including the step of transfecting a suitable cell with one or more gene constructs derived by internally deleting a nucleic acid encoding an influenza polymerase whereby said gene constructs are incapable of producing a functional polymerase but do not hinder packaging of gene segments into the viral particles. Further provided are virus particles obtained by such method, pharmaceutical compositions containing particles, and the use of such particles for the manufacture of a pharmaceutical composition directed at generating immunological protection against influenza.
