TRANSGENIC NON-HUMAN ANIMAL PRODUCING HETEROLOGOUS ANTIBODY

PROBLEM TO BE SOLVED: To provide a transgenic non-human animal producing heterologous antibodies and a method for producing human sequence antibodies which bind to human antigens with substantial affinity.

SOLUTION: A cell containing at least one artificial gene encoding at least a part of immunoglobulin polypeptides is provided. The cell produces a detectable amount of immunoglobulin bonding to a predetermined human antigen. The immunoglobulin polypeptides substantially have the same sequence as that of the immunoglobulin polypeptides secreted by hybridomas obtained from transgenic mice. The mouse includes a pair of homozygoses of functionally destroyed endogenous heavy chain allele, a pair of homozygotes of functionally destroyed endogenous light chain allele, at least one copy of a human immunoglobulin light chain transgene and at least one copy of a human immunoglobulin heavy chain transgene. The immunoglobulin binds to the human antigen with at least a 1.1×109M-1 affinity constant (Ka) in order to bind to the human antigen.
