A biological observation apparatus and a method thereof are provided to easily select a treatment technique about a tumor tissue by defining a part infiltrated by the tumor tissue inside a biological tissue. A sound wave generating part outputs a sound wave to a check object inside a biological tissue. A light generating part(21) irradiates a light to the check object. A light interference part emits interference light by intruding a light irradiated from the light generating part into a light reflected from a reaching region of the sound wave. A light detecting part(27) outputs an interference signal corresponding to the interference light emitted from the light interference part. A calculating part calculates an amount of frequency modulation of the emitted light based on the interference signal outputted from the light detecting part.
