Disclosed is a composition comprising: (a) a main species HER2 antibody that binds to domain II of HER2 and comprises a variable light amino acid sequence provided as SEQ ID NO: 4; and (b) the main species HER2 antibody further comprising an amino-terminal leader extension comprising VHS-, wherein from 5% to about 15% of the antibodies in the composition comprise the amino-terminal leader extension, as quantified by cation exchange analysis.
