<P>PROBLEM TO BE SOLVED: To provide a simple and efficient method of preparing a FVIII C2 protein using gene recombination technology wherein E. coli is used as a host. <P>SOLUTION: The simple and efficient method of preparing the above protein includes a process of adding urea into bacterial cells of recombinant E. coli wherein a polynucleotide including a C2 domain gene of blood coagulation factor VIII is introduced in an amount in a range of 1M to 3M relative to the intracellular soluble fraction present subsequent to the expression of the C2 domain protein of blood coagulation factor VIII, and a process of subsequently purifying the protein by performing metal chelate chromatography in the presence of urea in an amount in a range of 1M to 3M. <P>COPYRIGHT: (C)2009,JPO&INPIT |
