PROBLEM TO BE SOLVED: To provide new compounds which inhibit caspase-mediated cell apoptosis and inflammation. SOLUTION: This invention provides compounds of formula (Ia) or (Ib), wherein Z is oxygen or sulfur; R<SP>1</SP>is hydrogen, -CHN<SB>2</SB>, -R, -CH<SB>2</SB>OR, -CH<SB>2</SB>SR, or -CH<SB>2</SB>Y; Y is an electronegative leaving group; R<SP>2</SP>is CO<SB>2</SB>H, CH<SB>2</SB>CO<SB>2</SB>H, or esters, amides or isosteres thereof; R<SP>3</SP>, R<SP>4</SP>, R and R<SP>5</SP>are as described in the specification.
