PROBLEM TO BE SOLVED: To provide a biological desulfurization apparatus starting-up by depositing sulfur oxidation bacteria on a support filling layer while circulating a liquid or sludge containing microorganisms, with which the timing of stopping the circulation of the liquid or the sludge containing the microoraganisms and switching to sprinkling can be determined, and provide a start-up method thereof. SOLUTION: The biological desulfurization apparatus is provided with a biological reaction tank 1 into which a gas containing hydrogen sulfide gas is introduced, the support filling layers 2a, 2b which are arranged in the biological reaction tank and in which a support for depositing the microorganisms is filled, a means for supplying a gas containing oxygen to the biological reaction tank and two or more kinds of sprinkling mechanisms 3, 4 for sprinkling water necessary for the living substance to the upper part inside the biological reaction tank.
