Provided are mono-modified granulocyte colony-stimulating factor (G-CSF) by Y-type branched polyethylene glycol at the specific lysine position (Lys17) and the preparation method thereof. The pegylated G-CSF can be used for manufacture of pharmaceutical composition for treating neutropenia induced by infection, leukaemia and chemotherapy etc.
