PROBLEM TO BE SOLVED: To provide a method which efficiently acquires more peptide fragments in the case where peptide is fragmented by enzymatic digestion because the securement of the amount of the peptide fragments necessary for mass analysis is difficult in the case where the amount of peptide to be analyzed is small in the fragmentation of peptide by enzymatic digestion, and because the establishment of a method for enzymatically obtaining the peptide fragments in an amount related to the sensitivity of mass analysis even from a very small amount of peptide is desired. SOLUTION: This method is employed for hydrolyzing peptide in a buffer solution by protease, and the buffer solution includes a buffer solution containing at least a dipolar aprotic solvent.
