A composition for preventing and treating alcohol dependence using Puerariae Radix and Hoveniae Semen cum Fructus is provided to reduce the expression of POMC mRNA which is related to alcohol dependence. A composition for preventing and treating alcohol dependence comprises herb mixture extract as an active ingredient. The herb mixture extract is obtained with water, alcohol or their mixture solvent. The herb mixture contains each 2-30 weight% of Puerariae Radix, Puerariae Flos, Hoveniae Semen cum Fructus, Cyperi Rhizoma, the Alpinia officinarum, Chinese quince, Atractylodis Rhizoma alba, Ponciri Fructus, Aurantii nobilis Pericarpium, Zedoariae Rhizoma and Sophorae Radix.
