A method for preparing a peptide using usp2-cc is provided to quickly and easily produce heterologous peptide without the removal process of urea. A gene construct comprises a fusion partner, ubiquitin, and polynucleotide coding a fusion protein of heterogous peptide. A method for producing water soluble heterogous peptide comprises: a step of inducing an expression vector containing the gene construct and transforming; a step of culturing the transformant to induce the expression of recombinant fusion protein; a step of suspending the recombinant fusion protein in solution containing 2-4M urea; and a step of treating Usp2-cc in the suspension solution and cutting the fusion protein to obtain the heterogous peptide.
