A novel method for culturing a heterotrophic microalga and method for collecting a biomass are provided to suppress the reduction of readjustment or concentration of the heterotrophic microalga and increase the concentration of protein and chlorophyll of heterotrophic microalga. A method for culturing a heterotrophic microalga comprises: a step of culturing one or more heterotrophic microalga under the heterotrophic culture condition; a step of inoculating the culture medium; a step of culturing the heterotrophic microalga in a photobioreactor under the autotrophic culture condition; a step of adding a carbon source in a semi-discontinuous method; and a step of collecting biomass from the heterotrophic microalga through centrifugation.
