PROBLEM TO BE SOLVED: To provide a method for detecting the amount of luciferase in a sample, using light-emitting reaction of luciferin-luciferase, specifically, to provide a method for simultaneously detecting beetle-originated luciferase, having light-emission spectra characteristics with a light-emitting reagent containing Nonidet (R) P40 or polyoxyethylene (20) cetyl ether, and to provide a reagent therefor. SOLUTION: This method is for measuring the light emission of the luciferase in a cell culture solution that expresses one or more luciferases selected from among a group, consisting of green color light-emitting luciferase originating from Rhagophthalmus Ohbai, a variant-type light-emitting luciferase originated from Rhagophthalmus Ohbai, and red light-emitting luciferase originated from Phrixothrix hirtus, and comprises (a) a process for mixing cells with D-luciferin, adenosine triphosphate, a magnesium salt, and Nonidet (R) P40 or polyoxyethylene (20) cetyl ether as a surfactant; and (b) a process for measuring the light emission of the mixture solution.
