METHOD FOR PRODUCING MEMBRANE PROTEIN

It is intended to provide a method for producing a membrane protein folded into a native conformation or an active conformation in a lipid disc or a liposome. The method comprises the steps of: (a) preparing a cell-free protein synthesis reaction liquid containing a polynucleotide encoding a membrane protein, a steroidal surfactant, and a phospholipid, wherein the steroidal surfactant is contained at a concentration higher than the critical micelle concentration; (b) decreasing the concentration of the surfactant in the reaction liquid; and (c) synthesizing the membrane protein and at the same time, forming a lipid disc or a liposome integrated with the membrane protein (Fig. 5).
