发明名称 A KNOCK-OUT VECTOR CONSTRUCTED BY USING RFP REPORTER KNOCK-IN VECTOR AND METHODS FOR PREPARING THEREOF AND KNOCKING OUT GENE IN ANIMAL CELL
摘要 A knock-out vector prepared by using an RFP(red fluorescent protein) reporter knock-in vector is provided to express reporters more stably and effectively than an existing IRES(Internal Ribosome Entry Segment) system and knock out functions of a specific gene. An RFP(red fluorescent protein) reporter knock-in vector having a base sequence of SEQ ID NO:1 replaces initiation codon ATG of a gene to be knocked out with an ORF(Open Reading Frame) sequence containing initiation codon of GFP genes. The gene to be knocked out represents TIEG2(Transforming Growth Factor beta-inducible early gene 2). An existence of the knock-out is confirmed by measuring degree of fluorescence shown according to expression of RFP proteins.
