A chimeric mycobacterium tuberculosis gene vaccine is provided. The vaccine comprises Ag85a gene encoding a structural protein of mycobacterium tuberculosis as shown by sequence 1 and ESAT6 gene of mycobacterium tuberculosis as shown by sequence 2, wherein the ESAT6 gene is inserted into the sequence of Ag85a gene, and the Ag85a gene is inserted to eukaryotic expression vector pVAX1. A method for preparing the chimeric mycobacterium tuberculosis gene vaccine (plasmid) comprises the following steps: the PCR amplification of Ag85a gene; the construction of gene vaccine HG85 (plasmid); the PCR amplification of ESAT6 gene and the insertion of ESAT6 gene into plasmid HG85. The immunogenicity of the chimeric mycobacterium tuberculosis gene vaccine of the present invention is enhanced both in mice and monkey. The immunity effect of this vaccine is better than that of the single gene vaccine. |
