An enzyme-linked immunosorbent assay is provided to improve detection sensitivity for antigen within a biomaterial sample by integrating chemical signal amplification using minute particles with biochemical signal amplification using a cascade reaction. An enzyme-linked immunosorbent assay comprises the following steps of: collecting antigen using magnetic macroparticles wherein a specific primary antibody is fixed; treating the collected antigen using silica nanoparticles wherein a cascade reactive initiator and a specific secondarily antibody are fixed; treating precursor enzyme to be transformed to an active enzyme by the cascade reactive initiator and a substrate for specific signal formation; and measuring change of formation signal. The precursor enzyme indicates trypsinogen or Chymotrypsinogen.
