Provided is a method for separating a plant stem cell line derived from a cambium of herbaceous plant through a stable cultivation without a dedifferentiation process. A method for separating a plant stem cell line derived from a cambium of herbaceous plant comprises the following steps of: preparing a cambium contained storage root tissue of a herbaceous plant; cultivating the cambium contained storage root tissue in a culture medium including IAA(Indole-3-acetic acid) or IBA(Indole-3-butyric acid) to induce the plant stem cell line derived from a cambium of herbaceous plant; and collecting the induced plant stem cell line.
