PROBLEM TO BE SOLVED: To provide a microfluidic assembly, system, and method for manipulating fluid samples. SOLUTION: The assembly 98 includes an elastically deformable cover layer 104 and a less elastically deformable substrate 100. The method includes a process for deforming the substrate 100 via the cover layer 104 so that a new communication results in the assembly between the cover layer 104 and the substrate 100 and/or a new barrier wall is formed.
