摘要 An ion-exchange reagent comprising a layer of styrene-divinyl benzene copolymer ion-exchange resin coated upon, and at least partially polymerized upon, the surface of an inert extender of large superficial area is used for the separation of protein and polypeptide molecules from their solutions. In examples styrene and divinyl benzene are polymerized in the presence of benzoyl peroxide, methyl alcohol or chloroform, and a diatomaceous earth treated with dichlorodimethyl silane, and the coated extender sulphonated to form the ion-exchange resin. Other conventional functional radicals may be used. The composite resin is used to separate the proteins in a solution of insulin, or the peptides from a mixture prepared by the hydrolysis of insulin with crystallized trypsin.ALSO:Proteins and polypeptides are separated from solutions containing them e.g. insulin, and a mixture of peptides prepared from the B-chain of insulin by hydrolysis with crystallised trypsin, by adsorbing upon an ion-exchange reagent comprising an ion-exchange resin based on a styrene-divinyl benzene copolymer coated and at least partially polymerised on an inert extender of large superficial area e.g. diatomaceous earth. The separation is by chromatographic methods and a copolymer of styrene-divinyl benzene containing 5 to 15% of divinyl benzene is preferred for the separation of protein and 0,5 to 2,1% of divinyl benzene for the separation of polypeptides of MW 500 to 5,000. Examples describe the use of sulphonated copolymer coated upon diatomaceous earth for the treatment of insulin solutions of varying purity, and the mixture of peptides referred to above.ALSO:Insulin is separated from solutions containing same by adsorbing upon an ion-exchange reagent comprising an ion-exchange resin based on a styrene-divinyl benzene copolymer coated, and at least polymerized upon an inert extender of large superficial area e.g. diatomaceous earth. The separation is by chromatographic methods and a copolymer of styrene-divinyl benzene containing 5 to 15.1 of divinyl benzene is preferred for the separation of proteins and 0.5 to 2.1 of divinyl benzene for the separation of polypeptides of MW 500 to 5000. Examples describe the use of sulphonated copolymer coated upon diatomaceous earth for the treatment of insulin solutions of varying purity and the mixture of peptides referred to above.
