Methods and compositions for amplification of a target polynucleotide sequence by suppressing amplification of related sequences are provided. The method uses a polymerase lacking 5'-3' exonuclease activity and a non-extendable blocker oligonucleotide that hybridizes preferentially to a non-target sequence and downstream of a primer, thereby preventing template-dependent primer extension by the polymerase. |
