<P>PROBLEM TO BE SOLVED: To provide a method for base substitution in a double-stranded DNA using a DNA ligase which is an enzyme catalyzing phosphate binding of a DNA base. <P>SOLUTION: There provided is the method for the base substitution of the DNA comprising binding the double-stranded DNA (a) having a protruding end to a double-stranded DNA (b) having a protruding end complementary to the protruding end and an excessive base which is a kind different from the base present in the protruding end or a notched end of the (a) using the DNA ligase, thereby removing the base of the (a) present in a position corresponding to the excessive base and carrying out joining. <P>COPYRIGHT: (C)2009,JPO&INPIT |
