An apparatus for analyzing ethanol fuel ratio and determining fuel increase and an operation method thereof are provided to analyze the ethanol fuel ratio by using liquid molar fraction, fuel pressure, and fuel temperature and determine the increase of fuel amount. An operation method of an apparatus for analyzing ethanol fuel ratio and determining fuel increase comprises the steps of: standing until the newly filled fuel reaches the thermodynamic equilibrium state(S210); measuring the liquid molar fraction of the fuel; producing an operation index indicating volatility by using the pressure and temperature of the fuel when the measured molar fraction reveals that the fuel is gasoline, while producing the ethanol ratio in the fuel in case of gasoline and ethanol mixed fuel(S212); and determining a fuel increase factor by comparing the operation index or the ethanol rate with the standard value(S220).
