1355977 Spiro-dibenzocycloheptene-N-alkylisoindoline derivatives SANDOZ Ltd 9 July 1971 [13 July 1970 8 March 1971] 32277/71 Heading C2C Novel compounds of the Formula (I) are obtained by (a) selectively reducing the carbonyl group of the compound of Formula (II) in an inert organic solvent, and under an inert atmosphere, or (b) cyclizing the compound of Formula (III) with a mineral acid or with an organic acid or acid anhydride. Intermediates (II) may be obtained by cyclizing a compound of Formula (IV) with a mineral acid or with an organic acid or anhydride. Intermediates (III) may be obtained by reducing a compound of Formula IV, above, in an inert organic solvent and under an inert atmosphere. The intermediate IV may be obtained by condensing a compound of the Formula (V) with a compound of the Formula (VI) in an inert organic solvent and hydrolysing the resulting product. Pharmaceutical compositions having analgesic activity comprise the compound of Formula (I), or a pharmaceutically acceptable acid addition salt thereof, in association with a pharmaceutically acceptable diluent or carrier, in forms suitable for oral or parenteral administration. |
