PURPOSE:To isolate and purify phyllodulcin, by converting hydrangenol into its alkai salt with a (water-containing) lower alcohol extracted solution of leaves of hydrangea in a pH not less than 8, extracting a colored component, etc. with a weakly polar organic solvent in a pH not less than 11, removing it, followed by extracting phyllodulcin with a wealky polar organic solvent in a pH not more than 10. CONSTITUTION:In extracting and purifying phyllodulcin useful as a sweetener, perfume for tabacco, etc. from leaves of hydrangea, first the leaves of hydrangea is extracted with a lower alcohol, e.g., methanol, etc. or a water-containing lower alcohol to give an extracted solution, which is adjusted to a pH not less than 8 with an alkali aqueous solution and stirred at from normal temperature to about 50 deg.C for 30min to 6hr so that hydrangenol is converted into its alkali salt. The prepared solution is adjusted to a pH not less than 11 and extracted with a weakly polar organic solvent, e.g., chloroform, so that colored components and impurities are removed by the extraction. The prepared water-alcohol solution is adjusted to a pH not more than 10 and extracted with an organic solvent, e.g., chloroform, to give high-purity phyllodulcin in high yield. This substance is recrystallized from a lower alcohol to give an almost purified product. |
