PLASMID VECTOR COMPRISING YEAST AS HOST

摘要

PURPOSE:To raise manifestation efficiency of vector, by preparing a plasmid vector which contains a promoter and a terminator of gene of yeast pyruvate kinase, has 6.9Kbmol.wt. and specific yeast as a host. CONSTITUTION:EcoRI-XbaiDNA fragment of about 0.5Kb containing part of a promoter of a gene of yeast pyruvate kinase is obtained from chromosome DNA of Saccharomyces cerevisiae. The DNA fragment is inserted between BamHi site of plasmid pkY51 and EcoRI site to give plasmid vector pkY54 having 6.9Kbmol.wt. The plasmid vector is suitable with respect to function and antigenicity because N end of produced protein contains no amino acid sequence derived from yeast when an adventitious gene is inserted into the BamHI site.
