NEW MATERIAL:The human-originated physiologically active proteinous substance having the following properties. Molecular weight, 50,000+ or -5,000 (gel filtration, etc.); isoelectric point, 6.5+ or -1.0 (isoelectric electrophoresis); thermal stability, resistant to inactivation at 70 deg.C; physiological activity, exhibiting cell differentiation-inducing action to leukemia cell. USE:A differentiation-inducing agent for leukemia cell and a remedy for leukemia. PREPARATION:For example, a human-originated macrophage-like cell (e.g. peripheral blood monocyte) is cultured in a medium in the presence of a macrophage-activation substance such as phorbol ester and the supernatant liquid is collected and centrifuged to remove the cell scraps and obtain a solution containing a cell differentiation-inducing substance. The solution is subjected to biochemical separation operations such as concentration by ultrafiltration, dialysis and desalting, ion exchange chromatography, gel-filtration, electrophoresis, etc., to obtain the objective cell differentiation-inducing substance in high purity. |
