NEW MATERIAL:An optically active acyclonucleoside derivative expressed by formula 1 (R is heterocyclic base; X is halogen, tosyloxy, azido or amino). EXAMPLE:(S)-(+)-1-[(2'-Azido-1'-(hydroxymethyl)ethoxy)methyl]thymine. USE:Useful as a selective biosynthesis inhibitor for nucleic acid and therefore effectively used as a carcinostatic and antiviral agents. PREPARATION:An optically active compound expressed by formula 2 (Ac is acyl) is used as a starting raw material and hydroxyl group of the compound expressed by formula 2 is tosylated according to the reaction formulas (Ph is phenyl; Ts is tosyl). After deacylation, catalytic reduction is carried out to provide a compound expressed by formula 5. Two hydroxyl groups thereof are crosslinked with methylidene groups and then crosslinking is opened in the presence of a Lewis acid catalyst. The heterocyclic base having previously protected with trimethylsilyl group is then reacted therewith in the presence of a Lewis acid catalyst to afford a compound expressed by formula 8, which is subsequently deacylated, etc., to provide the compound expressed by formula 1. |
