PURPOSE:To enable utilization for waste water treatment by growing a microorganism in water containing salt and decomposing phenols at high temperatures under aerobic conditions. CONSTITUTION:Underground brine is added to a culture medium containing phenol, acclimatized and cultured at about 50 deg.C to provide high-temperature phenol-decomposing microorganism. The resultant decomposing microorganism is then repeatedly cultured in an artificial culture medium of prepared seawater and purified to afford TN501 strain (FERM P-11778), having the following properties and belonging to the family Rhizobiaceae. Microscopic shape; bacillus of 1X3mum size with motility without sporulation by culturing at 37 deg.C for 3 days and regular circular colonies with low convex circular protrusions and 1-1.5mm diameter by culturing in seawater agar culture medium for 24hr. Physiological properties; reducing nitrates and assimilating glucose without producing hydrogen sulfide and hydrolyzing starch. Having halotolerance to 7wt.% NaCl without any tolerance to 10wt.%. Susceptibility to antibiotic; negative to ampicillin and positive to chloramphenicol, etc. Decomposing properties of phenols; suitable at pH 5-11 and 20-65 deg.C at 0.2-7.0wt.% NaCl concentration, etc. |
