PROBLEM TO BE SOLVED: To provide an improved method for one-step RT-PCR (Reverse Transcriptase Polymerase Chain Reaction). SOLUTION: The invention relates to the method for performing the one-step RT-PCR for amplifying a target RNA comprising the steps (a) providing a sample which is supposed to contain the target RNA, (b) adding a reaction mixture comprising all reagents necessary to reverse transcribe the target RNA into cDNA and amplify at least a portion of the cDNA, (c) incubating the sample for a time interval of 0 seconds to 40 seconds at a temperature between 20°C and 65°C, and (d) subjecting the sample to multiple cycles of a thermocycling protocol wherein the temperature of the sample is varied between at least a first temperature between 90°C and 100°C and a second temperature between 50°C and 75°C.
