发明名称 A CRYSTALLIZED RECOMBINANT HEPARINASE I AND THE METHOD FOR CRYSTALIZING THE SAME
摘要 A crystallized recombinant heparinase I having heparin-neutralizing and angiogenesis-inhibiting effects is provided to facilitate X-ray crystallography and improve crystallinity, so that the three-dimensional structure of recombinant heparinase I is easily identified. A crystallized recombinant heparinase I produced from Bacteroides thetaiotaomicron VPI-5482 is prepared by using hanging-drop vapor diffusion method using reservoir solution containing precipitating agent and buffer solution, having pH 2.0-8.0 at 15-25 deg. C for 1 day to 1 week, wherein the recombinant heparinase I is prepared by fusing 6 histec fusions with a protein in the carboxyl terminal without deletion of amino acid residue in the amino terminal; and the precipitating agent is 1.0-2.4M of ammonium sulfate and the buffer solution is 1-5% citric acid solution.
