PROBLEM TO BE SOLVED: To provide a method for isolating and purifying a nucleic acid which has excellent isolating performance, cleaning efficiency, easy to process, and can be mass-produced and have substantially the same isolating performance, and to provide units for isolation and purification of the nucleic acid suitable for carrying out the method, and to improve passing rate of a lysate solution or cleaning solution by improving clogging in extracting operation while holding the high yield and high purity. SOLUTION: The method for extracting the nucleic acid uses a nucleic acid isolating and purifying cartridge including a container having two or more ports and housing solid materials through which a solution is passable, and the method shortens the passing time of the lysate solution and the cleaning solution and improves clogging in extracting operation. The process comprises following processes, (a) a process dispersing a biomaterial using a dispersing liquid, (b) a process eluting the nucleic acid by dissolving the biomaterial with a solution of a chaotropic salt or the like, (c) a process carrying out pipetting and stirring, (d) a process adding a water soluble organic solvent, (e) a process carrying out pipetting and stirring, (f) a process adsorbing the nucleic acid by a solid material, (g) a process washing impurities remaining, and (h) a process desorbing the absorbed RNA by a recovering solution.
